چکیده

A water soluble acidic heteropolysaccharide named wAF was isolated from Auricularia auricula-judae. It composed of a backbone of α(1→3)-linked D-mannopyranose residues. WAF fractions exhibited strong inhibition against Acinar cell carcinoma proliferation in vitro test. It significantly inhibited tumour growth in dose-dependent way in BALB/c mice. Moreover, the enhancement ratios of mice body weight for all wAF fractions were higher than that for 5-Fluorouracil, which is a chemotherapeutic anticancer agent. Fluorescence microscopy and tumour tissues cell morphology observation indicated that wAF3 fraction induced apoptosis of tumour cell. Immunohistochemistry for apoptosis-related proteins Bax and Bcl-2 expression revealed that wAF3 induced S-180 tumour cell apoptosis by up-regulation of Bax and down-regulation of Bcl2. These findings confirmed that the wAF polysaccharide had good antitumour activity and could be considered as a potential antitumor agent. Comparative Antitumor Activity of Different Solvent Fractions from an Auricularia auricula-judae Ethanol Extract in P388D1 and Sarcoma 180 Cells

The objective of this study was to evaluate and compare the antitumor activity of different solvent fractions (ethanol, dichloromethane, ethyl acetate, butanol and water) of the Auricularia auricula-judae 70% ethanol extract on the P388D1 macrophage and sarcoma 180 cells. A dose-dependent antitumor activity of each solvent fraction (from 0.01 mg/ml to 0.3 mg/ml) was shown against both cell type...

Development of Detection Methods for Cellulolytic Activity of Auricularia auricula-judae

To obtain basic information on the detection of cellulolytic activity in Auricularia auricula-judae, the influences of dye reagent, pH, and temperature were assessed. Chromogenic dye (congo red, phenol red, remazol brilliant blue, and trypan blue) was individually incorporated into a medium containing either carboxymethyl-cellulose, Avicel, or D-cellobiose as a polysaccharide carbon substrate. ...

Anti-inflammatory Activity of Dichloromethane Extract of Auricularia auricula-judae in RAW264.7 Cells

The present study investigated the anti-inflammatory effects of dichloromethane extract of Auricularia auricula-judae. Dichloromethane extract of Auricularia auricula-judae inhibited Lipopolysaccharide (LPS) -induced nitric oxide (NO) production significantly in a dose-dependent manner in the concentration ≥10 μg/ml (p < 0.05) . Furthermore, RT-PCR results of this study indicated that the extra...

Hypolipidemic Effects of Biopolymers Extracted from Culture Broth, Mycelia, and Fruiting Bodies of Auricularia auricula-judae in Dietary-induced Hyperlipidemic Rats

Hypolipidemic effect of biopolymers extracted from culture broth (CP), mycelia (MP), and fruiting bodies (FP) of Auricularia auricula-judae was investigated in dietary-induced hyperlipidemic rats. The experimental animals were administrated (100 mg/kg body weight) with different biopolymers, daily for 4 weeks. Hypolipidemic effects were achieved in all the experimental groups, however, FP was p...

Structural Characterization of Melanin Pigments from Commercial Preparations of the Edible Mushroom Auricularia auricula

Many of the most widely consumed edible mushrooms are pigmented, and these have been associated with some beneficial health effects. Nevertheless, the majority of the reported compounds associated with these desirable properties are nonpigmented. We have previously reported that melanin pigment from the edible mushroom Auricularia auricula can protect mice against ionizing radiation, although n...
